Two goals in nine minutes towards the end of the first half from Pedro Goncalves put Sporting in control and after Dortmund lost Emre Can to a straight red card on 74 minutes, Pedro Porro made it three on the rebound as Goncalves saw his penalty saved and missed out on a hat-trick.
Donyell Malen's stoppage time strike was scant consolation for the visitors.
The result puts Sporting on nine points from five games, six behind Ajax, who maintained their 100 per cent record with a 2-1 win at Besiktas.
Dortmund have six points, but cannot now overhaul Sporting regardless of what happens in the final round of matches as Sporting have the superior head-to-head record and will play in the knockout stage for the first time since the 2008-09 season.
